Streptomyces-derived natural products are an important source of lead compounds for drug discovery. Traditional methods for mining these compounds are costly, time-consuming, and labor-intensive. To address these challenges, we applied NP-PRESS, a two-stage metabolome refining pipeline, to Streptomyces speibonae W307, isolated from a mangrove environment. We conducted a thorough analysis of molecular features using NP-PRESS, leading to the identification of three natural products named strepyrazinones A, B, and C, two of which possess new structures. These compounds were characterized using high-resolution MS and NMR, with their configurations determined through ECD calculations. Additionally, we proposed their corresponding biosynthetic gene cluster and putative biosynthetic pathways.
