Osteosarcoma (OS) is a common primary bone malignancy predominantly affecting adolescents. The bone microenvironment is crucial in modulating the initiation and progression of OS. OS cells transduce mechanical signals into biochemical signals, thereby regulating cellular behavior. A comprehensive examination of the complexities of mechanotransduction in OS strengthens the theoretical framework for understanding its regulatory mechanisms. Existing theoretical frameworks often fail to integrate biomechanical cues with epigenetic regulation, which is essential for a comprehensive understanding of OS pathogenesis. This review highlights the aberrant mechanotransduction in OS cells, contrasting with physiological bone remodeling, examines key signaling molecules involved, and summarizes potential therapeutic strategies targeting mechanotransduction. These insights provide a foundation for developing innovative and targeted therapies for OS.
